Analysis of Zerumbone Use Densitometry

ABSTRACT

The research done analysis of Zerumbone amount in lempuyang gajah (Zingiber zerumbet(L).J.E Smith) rhizome with extraction method and solvent variety. This study aims to know influence extraction method and solvent variety about zerumbone in Zingiber zerumbet rhizome. Spot color, Rf and spectrum for qualitative analysis then Densitometrically use TLC plate silica gel F254  with mobile phase n-Hexane - ethyl acetate (9:1) at maximum absorbance wavelength 259 nm for quantitative analysis. Analysis with variety  extraction method according soxhlet, reflux, maceration using methanol and water distillation acquired that reflux have maximum zerumbone content (25,08%) in extract. Analisis with solvent variety according n-Hexane, Ethanol 70%, and Aquadest using reflux method show that n-Hexane have maximum zerumbone content (53,47%) in extract.

        Telah dilakukan penelitian analisis kadar Zerumbone pada rimpang lempuyang gajah (Zingiber zerumbet (L).J.E.Smith) dengan berbagai metode ekstraksi dan variasi cairan penyari . Penelitian ini bertujuan untuk mengetahui pengaruh metode ekstraksi dan cairan penyari terhadap kadar zerumbone pada rimpang lempuyang gajah. Analisis kualitatif dilakukan dengan melihat warna noda, Rf, dan spektrum, kemudian dilanjutkan dengan analisis kuantitatif secara Densitometri menggunakan lempeng KLT silica gel F254 dengan eluen n-Heksan – etil asetat (9:1) pada panjang gelombang 259 nm. Analisis dengan berbagai metode ekstraksi secara soxhlet, reflux, maserasi menggunakan penyari m]ethanol dan destilasi air diperoleh bahwa metode reflux mengandung zerumbone sebanyak (25,08 %) dalam ekstrak. Dan analisis dengan variasi cairan penyari n-Heksan, Etanol 70%, dan Aquadest menggunakan metode reflux menunjukkan bahwa cairan penyari n-Heksan mengandung zerumbone sebanyak (53,47%) dalam ekstrak
